Are you using Windows Indexing or Everything for your search?
Flow uses Windows Search or Everything to do the actual file searching. Activate Flow and type in Settings. Then go to Plugins - Explorer and check the options there.

Are you using Everything or Windows Index as your search engine (can be found in the plugin settings)?
Why are you using a space for the filespec after the folder? `C:\downloads\*.jpg` will find those files
