No a blastn mode is not supported at this time.
No I don't think that would work well in practice without adapting a lot of other things in the algorithm.
Initilization needs to be run if you want to use the `--multiprocessing` feature, otherwise its not needed.
You need to include the path to the `diamond` file e.g. `./diamond blastx ...`.
You seem to be using a Mac but have downloaded the linux version. On Mac you have to install either using bioconda, homebrew or compile from source (see the Wiki).
It means the file does not exist. Make sure you have the correct path and file name.
It's not possible to write the DAA format to stdout, since this format is not being written linearly, but the program needs to seek within the file.
Please let me know the exact error message you are getting.
