Fiducial Markers Trajectory Smoothing Factor Error

Open babim2 opened this issue 8 years ago • 1 comments

Inputting a value of 0 for the trajectory smoothing factor while correcting for dirft using fiducial markers causes the following error to appear: Smoothing_Factor_Error.txt

The data set is acquired from the analysis of a single channel of fiducial markers. The frame is cropped to a single marker and SM-localization is conducted by ThunderSTORM. All localizations match the marker position and only a single localization is observed per frame. Attempting to correct for drift using the fiducial marker yields the error when using the following settings: Max distance: 40 nm Min marker visibility ratio: 0.1 Trajectory smoothing factor: 0

Thanks for all your help! ThunderSTORM has been an integral part of my research!

I have found that there is a minimum value for the smoothing factor below which there will be an error. It depends on the number of substacks (i.e. more substack means you can go lower). My guess is that the minimum value correspond to no smoothing. Maybe normalizing the factor between 0 and 1 whatever the substack number would be better?
