Minimum number of particles and symmetry

Hi @zhonge,

Hope you are well. There are a couple of questions that I did not get answer to in the paper. Could you please address them?

I ran CryoDRGN with ~1000 particles and ~5000 particles respectively from a dataset D1. However, none of them gave any sensible reconstruction.

Q1. Is there any minimum number of particles required to do heterogeneous reconstruction with CryoDRGN for any given dataset ? Q2. Regarding hetero reconstruction for symmetric particles, is it a necessity to do C1 consensus refinement for CryoDRGN? In my experience, C1 consensus refinement for symmetric particles usually does not generate a good 3D refined model. Q3. In case we generate, Initial model and 3D refined model both with a different symmetry than C1, would CryoDRGN not give sensible results?

I look forward to your response.

Thanks and Regards, Deepesh
