If you are using some other username instead of "pi" you will get the error. Change your username in /home/pi/env/bin/python from pi to your actual username.
Its difficult to says whats gone wrong without actually seeing the error. I need to look at the terminal contents etc.
Can you manually run the startsample.sh and post the terminal contents ? Have you authorized the alexa ?
Without running service installer if you give systemd commands it will return with service not found only. It needs to be done only after authorizing Alexa. I have mentioned that...

You have not given the commands according to the syntax that i have given. Please go through the README doc and the details about customization.
Have you input the spotify user-id in the config.yaml ?
I do not support Bluetooth speakers. It's up to the user to get the audio config done.
