My favourite features are:

• spell <word> for quick spell checking
• Integrated clipboard and snippet manager
• next for skipping to the next track
• empty for emptying the trash
• open <dir> for opening a directory
• Quick opening/switching of applications
• Quick internet searching

If I sit down at a Mac that doesn't have Alfred installed, I have to remember how to open things now. It is the most useful tool that I have ever installed, I think.
