Updates for terms GO:0030283, GO:0047045, GO:0047035 and GO:0035410
- Based on the description of the EC numbers, GO:0030283 and GO:0047035 appear to have a broad range of substrates which is not obvious from their name and definition. Could the following changes be made to reflect their broad specificity?
For
id: GO:0030283 name: testosterone dehydrogenase [NAD(P)+] activity def: "Catalysis of the reaction: testosterone + NAD(P)+ = androst-4-ene-3,17-dione + NAD(P)H + H+." [EC:1.1.1.51]
EC 1.1.1.51 Accepted name: 3(or 17)β-hydroxysteroid dehydrogenase Reaction: testosterone + NAD(P)+ = androstenedione + NAD(P)H + H+ Glossary: androstenedione = androst-4-ene-3,17-dione Systematic name: 3(or 17)β-hydroxysteroid:NAD(P)+ oxidoreductase Comments: Also acts on other 3β- or 17β-hydroxysteroids.
Proposed changes for name and def: id: GO:0030283 name: 3(or 17)β-hydroxysteroid dehydrogenase [NAD(P)+] activity def: "Catalysis of the reaction: testosterone + NAD(P)+ = androst-4-ene-3,17-dione + NAD(P)H + H+. Also acts on other 3β- or 17β-hydroxysteroids."
For
id: GO:0047035 name: testosterone dehydrogenase (NAD+) activity def: "Catalysis of the reaction: testosterone + NAD+ = androst-4-ene-3,17-dione + NADH." [RHEA:14929]
EC 1.1.1.64 Accepted name: testosterone 17β-dehydrogenase (NADP+) Reaction: testosterone + NADP+ = androstenedione + NADPH + H+ Glossary: androstenedione = androst-4-ene-3,17-dione Comments: Also oxidizes 3-hydroxyhexobarbital to 3-oxohexobarbital.
Proposed changes for name and def: id: GO:0047035 name: 3α(17β)-hydroxysteroid dehydrogenase (NAD+) activity def: "Catalysis of the reaction: testosterone + NAD+ = androst-4-ene-3,17-dione + NADH. Also acts on other 17β-hydroxysteroids and on the 3α-hydroxy group of pregnanes and bile acids."
- Currently GO:0030283 and GO:0047045 are at the same levels in the ontology.
Based on the reaction could GO:0047045 become a child of GO:0030283?
id: GO:0030283 name: testosterone dehydrogenase [NAD(P)+] activity def: "Catalysis of the reaction: testosterone + NAD(P)+ = androst-4-ene-3,17-dione + NAD(P)H + H+." [EC:1.1.1.51]
id: GO:0047045 name: testosterone 17-beta-dehydrogenase (NADP+) activity def: "Catalysis of the reaction: NADP+ + testosterone = NADPH + H+ + androst-4-ene-3,17-dione." [EC:1.1.1.64, MetaCyc:1.1.1.64-RXN]
- The reaction described in GO:0035410 is covered by the broad GO term GO:0030283 (see above); they share the same EC Xref. Would it make sense to merge GO:0035410 with GO:0030283?
id: GO:0035410 name: dihydrotestosterone 17-beta-dehydrogenase activity namespace: molecular_function def: "Catalysis of the reaction: 5alpha-dihydrotestosterone + NAD+ = 5alpha-androstane-3,17-dione + NADH." [GOC:ecd, PMID:4152755] xref: EC:1.1.1.51 is_a: GO:0033764 ! steroid dehydrogenase activity, acting on the CH-OH group of donors, NAD or NADP as acceptor
