Once installed can we update to Big Sur?

It might be possible. It's better to do this in VMware workstation instead of player (you will need a license key though) so you can take snapshots and if the upgrade fails you can always revert back to the snapshot. Otherwise you'll have to reinstall.

Do not update you virtual machine. The AMD kernel is only built for 10.15.0 and you will break your virtual machine if you update it.
