the number of samples
Hello, I have questions about the samples that may impact the assembly result.
Since my sample is relatively small, I had to pool them up for HiFi sequencing to extract enough DNA. However, I also discovered that my sample has a high heterozygous genome.
So the question is is it okay for me to pool 10 same species to generate Hifi data and assemble it?
P.S. the sequencing depth might be X25

I have no idea about this, but I heard some assemblies of the Darwin Tree of Life project were generated by mixing different samples. Probably you can ask for some suggestions from them?
