Fixed: issue was related to nginx + uwsgi... had to set buffer-size=65535 for my uwsgi config.

Correction, this just does not seem to work in win32 at all.
Finally figured out a solution for win32. The va_list (...) aspect of the libusb_set_option is not really supported by ctypes. Not sure why this works on 64bit but 32bit definitely...
I considered that but since its not a libusb bug I didnt think they would care. The bug is in ctypes.
