Play Hotkey not functioning, while all other hotkeys function perfectly

Good evening,

My play hotkey doesn't work. I don't know why. All the other ones work. My understanding was that I could have all my macros off (while keeping the window open on my computer) and click the Play hotkey, and it would then play the macro. Is that not what it does? I believe it is an issue on the program's end.
