12V HV enable pulse issue

This programmer https://www.tindie.com/products/leonerd/avr-updi-programmer-with-12v/ is implementing a 12V HV enable pulse, by RTS handshaking line being asserted when the serial port is opened. But, there is something not triggering that to happen. I would be great if this could be figured out, the programmer works fine, when the devices are not locked.
