I'm getting the same error. I have pngquant installed. I try the following:

import pngquant
pngquant.config('/path_to_pngquant_dir/pngquant')

I can run pngquant from the command line with /path_to_pngquant_dir/pngquant, but still get the ValueError in the python script.
