I would like to be able to customise the response of the bot on Mastodon.

A template/format string I can use in the config to customise the response.

None besides using the default.

I'm not sure how you'd template this out tho 🤔 In this text I should be able to place some "variables" corresponding to what I expect to come out in the reply. E.g. "$ia" for an internet archive link?

Do you mean that customising the response text "URL no found" when no URL is archived?

Not a bad idea, this allows more flexibility in setting up messages to respond to. I can't say at the moment how much this is needed, and how to set the message length limits, which I understand are different for different instances of Mastodon.
